Abstract

​Regulatory T cells (Tregs) suppress other immune cells and are critical mediators of peripheral self-tolerance, preventing autoimmune disease but hampering tumor rejection. Therapeutic manipulation of Tregs is subject to numerous clinical investigations with promising outcomes in first in-man trials of adoptive Treg transfer. The number of naturally occurring Tregs (nTregs) is minute, encouraging the complementary approach of inducing Tregs (iTregs) from naive T cells. Mouse studies exemplify the importance of peripherally induced Tregs along with applicability of iTreg transfer in disease models.
